- Abstract
- In the growing field of Nitric oxide(NO)-based therapy, there is a need to specifically determine the toxicity of NO since many medical applications are based on the anti-cancer effect of NO including metastatic cancer. Before the in vivo application, in vitro cell experiments are required to achieve a detailed knowledge of toxicity.
In this study, we show that cytotoxicity of NO, inspected by cellular experimental techniques is highly due to the formation of ROS-mediated ONOO- production. To further investigation, NO-induced cellular toxicity is dependent on ROS concentration in various cancer cell lines. The method is validated by conventional cytotoxicity testing and co-tested by fluorescence to visualize changes in the concentration of intracellular ROS and to determine the production of ONOO-.
